چکیده

Introduction NLRP3 has a pivotal role in nucleating the inflammasome, a cytoplasmic multiprotein complex that mediates the maturation of the proinflammatory cytokine interleukin1b (IL-1b) by activating caspase-1. Mutations in the gene encoding NLRP3 cause a spectrum of autoinflammatory disease, the cryopyrin-associated periodic syndromes (CAPS). It has been reported that generation of reactive oxygen species (ROS) is a major NLRP3 inflammasomeactivating factor. However, the molecular mechanism by which a change in cellular redox state leads to NLRP3 inflammasome activation has not been elucidated. Here we show that cofilin-1, a redox sensitive actin binding protein, is involved in NLRP3 inflammasome activation.

P 106: Effects of Dimethyl Sulfoxide on NLRP3 Inflammasome and Alzheimer\'s Disease

Alzheimer's disease (AD), the most ordinary form of dementia and extracellular accumulation of Amyloid-β (Aβ) in senile plaques, is an important and a main event in the pathogenesis of AD. Deposition of Aβ Peptide initiates a spectrum of cellular responses that are interposed by the resident neuroimmune cells of the brain, the microglia. Recently, a novel inflammasome signaling&n...

Hemin Induces the Activation of NLRP3 Inflammasome in N9 Microglial Cells

Background: Hemin is an important sterile component that induces a neuroinflammatory response after intracerebral hemorrhage, in which NLRP3 inflammasome activation has also proved to be involved. Although microglial activation acts as a key contributor in the neuroinflammatory response, the relationship between hemin and NLRP3 in microglia remains poorly understood. Objective: To investigate w...

NLRP3 inflammasome: From a danger signal sensor to a regulatory node of oxidative stress and inflammatory diseases

IL-1β production is critically regulated by cytosolic molecular complexes, termed inflammasomes. Different inflammasome complexes have been described to date. While all inflammasomes recognize certain pathogens, it is the distinctive feature of NLRP3 inflammasome to be activated by many and diverse stimuli making NLRP3 the most versatile, and importantly also the most clinically implicated infl...

Nuclear factor E2-related factor-2 (Nrf2) is required for NLRP3 and AIM2 inflammasome activation.

Despite the number of extensive studies on the immune function and signaling of inflammasomes in various diseases, the activating mechanism of inflammasome, especially the NLRP3 inflammasome, is not fully understood. Nuclear factor E2-related Factor-2 (Nrf2), a key transcription factor that regulates cellular redox homeostasis, has been reported to play both protective and pathogenic roles depe...

The bacterial pigment pyocyanin inhibits the NLRP3 inflammasome through intracellular reactive oxygen and nitrogen species

Inflammasomes are cytosolic complexes that mature and secrete the inflammatory cytokines interleukin 1β (IL-1β) and IL-18 and induce pyroptosis. The NLRP3 (NACHT, LRR, and PYD domains-containing protein 3) inflammasome detects many pathogen- and danger-associated molecular patterns, and reactive oxygen species (ROS)/reactive nitrogen species (RNS) have been implicated in its activation. The phe...
